Development of encapsulated organic solvent resistant multienzyme complexes for multiple step reactions

Project Overview
In general, small molecule biosynthesis requires multiple sequentially coupled enzyme reactions. Since many substrates and/or products are only partially soluble in water and most enzymes are less active or even inactive in organic solvents, cell free synthesis on a technical scale is complicated. The goal of this project is the development of synthetic microcapsules that contain a cascade of enzymes selected for organic solvent resistance by a mutagenic screening procedure. These enzyme should be able to catalyze reactions from a specific pathway in an aqueous mileau containing organic solvent. The efficiency of the semi-synthetic enzyme cascades in either the absence or presence of microcapsules will be analyzed by mathematical modelling of the intermediates produced by a given reaction sequence. Therefore, intermediates of the pathway will be measured using ion chromatography or HPLC coupled with mass spectrometric analysis. The modelling approach should allow the identification of possible bottlenecks which negatively affect the efficiency of the enzyme reactions in a closed environment.
